Figure 2: Pull-Down Assays Using TRAMPE Engineered Proteins

All human proteins were produced by cell-free expression and labeled with the BODIPY-FL fluorophore using Ambergen's TRAMPE technology. The "bait" proteins were immobilized on beads using an antibody to their common HSV epitope tag and the beads then treated with MDM2 protein. As expected, only the p53 "bait" pulls-down the MDM2, while the GST A2 negative control "bait" does not, in agreement with the known p53-MDM2 interaction. The "MDM2 Standard" simply shows the position of the labeled MDM2 on the gel. The gel was scanned in a fluorescence imager.
